ПААГЭ[греч. poly — много, лат. acri — едкий, англ. amino — группа NH2, от ammonia — аммиак, от лат. sol ammoniacus — соль Аммона, нашатырь и греч. eidos — вид; лат. gelare — застывать; греч. elektron — смола, янтарь и phoresis — несение, перенесение]широко используемый электрофоретический метод разделения макромолекул (РНК, ДНК, белков) с помощью полиакриламидного геля (см. полиакриламидный гель). Этот метод используется при секвенировании ДНК, определения гомогенности белковых препаратов и др. Метод основан на свойстве заряженных частиц (молекул) перемещаться под действием электрического поля. При анализе белков скорость их миграции в геле зависит от трех параметров: величины молекул, формы молекул и суммарного заряда. Поэтому предварительно белки денатурируют с помощью детергента додецилсульфата натрия (см. додецилсульфат натрия) с тем, чтобы скорость миграции зависела только от молекулярной массы. Под действием ДСН олигомерные белки диссоциируют на субъединицы и денатурируют (ПААГЭ в присутствии ДСН, SDS-PAGE).
